You have now entered all of the information in VISSIM necessary to run the simulation. You can set your simulation preferences by going to Simulation -> Parameters… Once all of your settings are to your liking, click the Play button to start the simulation. After a moment of loading the simulations will generate vehicles and your traffic signal should cycle through each phase as programmed.
